- PublicationAphrodisiac property and anti tumor agent of eurycoma longifolia jack (Tongkat Ali)(2012)Zakaria, YusmazuraEurycoma longifolia Jack, identified by its local name as Tongkat Ali is commonly found along the hilly jungle slopes of Malaysia. It has gained notoriety as a male aphrodisiac among malaysian since it is reputed to increase male sexual. In this present study, sexual attractiveness test showed that E. longifolia increased the sexual behaviour with regard to the lesser time taken for the male to copulate the estrus female. It was showed that E. longifolia aqueous extract increased the sexual activities in a dose and time dependent manner. The aqueous extract of E. longifolia also increased sexual responsiveness of female rats by decreased the time taken for the female rats to copulate towards the male rats. This study provides evidence that the aqueous extract of E. longifolia acts specifically on sex hormones, neurotransmitter nitric oxide (NO) in penile erection and also phosphodiesterase enzymes (PDEs). The extract increased the levels of testosterone in congruence with DHT in male rats. The highest level of testosterone was observed when the extract at 90mg/kg was used. At the highest concentration used in this experiment (150mg/kg) testosterone levels ‘slogged off. In treated female rats, aqueous extract of E. longifolia also regulated the testosterone and DHT levels including estrogen and progesterone. E. longifolia was also found to maintain penile erection. This extract acts via NO and PDEs. It was increased nitric oxide synthase (NOS) expression to form NO and inhibited PDE3, PDE4 and PDE5. The antiproliferation assay showed the extract inhibited cell proliferation towards human malignant melanoma cell (HM3KO), human cervical cancer cell (Hela), human liver cancer cell (HepG2) and human ovarian carcinoma cell (CaOVs). The extracts did not inhibit the cell proliferation for both normal cell lines used, human normal skin cell (CCD11114sk), and human normal liver cells, Chang’s liver. Eurycomanone is a compound found in E. longifolia. Eurycomanone significantly increased apoptosis in HepG2 cells (3.8±0.12 pg/ml) and showed less toxicity towards both normal liver cells, Chang’s liver (17±0.15 pg/ml) and WLR-68 (20±0.22 pg/ml) as compared to tamoxifen (1.4±0.31 p.g/ml) and vinblastine sulfate (4.2±0.37 pg/ml). The characteristics of apoptosis including chromatin condensation, DNA fragmentation and apoptotic bodies were found following eurycomanone treatment. The apoptotic process triggered by eurycomanone involved the up-regulation of p53 tumor suppressor protein. The up-regulation of p53 was followed by the increasing of pro-apoptotic Bax and decreasing of anti-apoptotic Bcl-2. The increased of cytochrome C levels in cytosol also resulted in induction of apoptosis.

- PublicationEstablishment of rapid detection of Haemophilus influenzae type b by isothermal amplification of bexA gene.(2015)Siong, Leow CheeHaemophilus influenzae type b (Hib) is a common cause of childhood meningitis that leads to a significant morbidity and mortality in both developing countries and underdeveloped countries, where the successful Hib conjugate vaccine is not accessible. A rapid and costeffective diagnostic method was highly demanded in resource-limited laboratory. Hence, the aim of this study is to establish a rapid detection method for H. influenzae type b by loopmediated isothermal amplification (LAMP) of bexA gene. A LAMP primer set was specifically designed to target bex\ gene. An in-house LAMP reaction was established and optimized using extracted genomic DNA of Haemophilus influenzae ATCC 10211. Subsequently, both LAMP reagents and reaction conditions were optimized and the optimized LAMP assay was subjected to analytical specificity and sensitivity evaluation. PCR reaction was conducted by using outer primers to provide comparison in term of analytical sensitivity. The analytical specificity was evaluated with thirty non-haemophilus strains which showed 100% specificity for bexX gene identification. The limit of detection (LOD) for LAMP assay was determined at 2 pg of DNA per reaction and as low as 2 colonies-forming units (CFU) per reaction. This indicates that the detection limit of LAMP by CFU per reaction was 100 fold lower than the detection limit of PCR. Based on the result obtained, LAMP has the potential to become an excellent diagnostic tool capable to produce a rapid, highly sensitive and specific result in a single temperature condition.
- PublicationFamily planning: attitudes and practices among postnatal women and their husbands in obstetric and gynecology clinic, Hospital Universiti Sains Malaysia (HUSM).(2009-04)Baser, Nor IdzzianaLack of awareness towards family planning among men and women is an issue needed to take into account. This quantitative study was conducted to identify the relationship between attitudes and practice of family planning among men and women in Obstetric and Gynecology Clinic, HUSM. This research is a cross-sectional, descriptive study using questionnaire. The sample is 65 men and women selected from Obstetric and Gynecology Clinic, HUSM. Self-administer questionnaire was utilized and the questionnaire consists of three sections. Section A consists of respondent's demographic characteristics, Section B consists of 26 attitudinal statements and Section C consists of one question regarding contraceptives practice. Data was analyzed by Chi-square test from Statistical Package for Social Science (SPSS) version 12 software. More than half of the men and women had positive attitudes towards family planning, but yet only minority of them uses contraceptives method. These quantitative findings contrast with results from other research in other countries. Educated men and women's tend to practice family planning due to their knowledge to the use of contraceptives. Men and women who wanted more children were reluctant to practice family planning. Understanding family planning thoroughly is the key concept in practicing it. Throughout this study, sociocultural and traditional norms give a huge impact on practice of family planning. Therefore, intervention efforts should address these issues and mold action plans appropriate to the situation.
- PublicationIsolation of adipose stem cells from human skin adipose tissue.(2009)Traditionally, stem cells are isolated from bone marrow. These cells have the potency to develop into various types of cells. Thus, mesenchymal stem cells from bone marrow can be used in treating the diseases that need cells therapies or regenerative of cells. Much of the procedures that involve the aspiration of stem cells from bone marrow were invasive and painful. Thus, many researches try to find the alternative source of stem cells. Adipose tissue which is ubiquitous in human body and can be easily isolated promise as a novel source for tissue engineering and regenerative medicine. When compared with bone marrow stem cells, adipose-derived stem cells may develop into adipocytes, chondrocytes, osteocytes and myocytes in vitro. In this research project, adipose tissues from humans were collected and isolated in vitro. The isolation procedures involved centrifugation, filtration and culturing with DMEM: F-12 (1:1), FBS and PenStrep. Adipose stem cells were isolated from human subcutaneous adipose tissue, and appeared as fibroblast-like morphology observed under inverted phasecontrast microscope.
- PublicationMaternal anxiety during universal newborn hearing screening at Hospital Universiti Sains Malaysia.(2009)Kamaruddin, Noor RafidahThe purpose of this study was to investigate the maternal anxiety during Universal Newborn Hearing Screening (UNHS) at Hospital Universiti Sains Malaysia (HUSM). A sample of 50 mothers who had given birth at HUSM was surveyed using a set of questionnaire. The instruments were based upon demographic profile, knowledge about hearing screening and Beck Anxiety Inventory (BAI). BAI is a set of questionnaire used to assess the level of maternal anxiety. The data was coded and analyzed using Wilcoxon signed Rank Test of SPSS version 11.5. The findings indicated that majority of the mothers were having mild anxiety during the first screening and before they undergone the second screening. The result also showed that there was significant difference of maternal anxiety level between the first screenings and before the second screening. There was also significant difference in the level of maternal anxiety between mothers whose babies failed the first screening and passed the second screening. Even though both results showed that there was a significant reduction in the level of maternal anxiety, but some of them are still in anxiety state. Therefore, actions need to be taken to improve the failure rate in UNHS.
- PublicationOptimization of the differential pulse anodic stripping voltammetry technique for determination of cadmium in Pengkalan Chepa River, Kelantan.(2009)Cadmium is one of heavy metal that can cause river water pollution by waste from factories. Common exposure of human or animal towards cadmium will cause toxicity. In determination of cadmium, voltammetric technique was used since it is highly sensitive, low cost and low limit of detection. This technique was used for qualitative and quantitative determination of cadmium in water samples. Samples were collected from five rivers; Sungai Alor B, Sungai Alor Lintah, Sungai Keladi, Sungai Pengkalan Chepa and Sungai Pengkalan Chepa 2 which are tributaries of main river, Pengkalan Chepa River at different depth (surface and bottom). Differential Pulse Anodic Stripping Voltammetry (DPSA V) technique was carried out using three electrodes, hanging mercury drop electrode (HMDE) act as working electrode, Ag/ AgCIIKCl saturated act as reference electrode and platinum electrode act as auxiliary electrode. The supporting electrolyte used was acetate buffer (1.38 M, pH 4.6). The optimum voltammetric parameters to determine cadmium in river water were initial potential (Ei) -1 V, final potential (Er) -0.2 V, scan rate (v) 5 mV/s, voltage step 0.005 V, voltage step time 1 s, deposition potential (Eacc) -1 V, deposition time (tacc) 60s and equilibrium time (teq) 1000 s. Using this optimized parameters, peak potential (Ep) for cadmium standard was found at -0.632 V. The linear range of cadmium was observed over the concentration range 0.26 ppm to 1.32 ppm with correlation coefficient (r) 0.993, standard deviation (SD) 0.500, sensitivity 10.227 nA/ppm and limit of detection 147 ppb. The method had been successfully applied for determination of cadmium in real samples. The results showed that cadmium was absent in all the samples.

- PublicationPhysical fitness of taekwondo athletes.(2009-04)A.Rashid, Noorul HusnaThe purpose of this study was to assess physical frtness in young Kelantanese taekwondo-in. The participants were eight male (18.63 ± 1.92 yrs, 168.65 + 7.36 em, 68.29 ± 20.69 kg) and ten female (18.10 ± 1.37 yrs, 158.22 ± 4.11 em, 59.72 ± 10.03 kg) taekwondo athletes. Weight, height and body composition were the anthropometric measurement that were assessed according to ISAK guidelines. The physical fitness characteristics that were evaluated included aerobic endurance by the PACER test, muscular strength and endurance by sit-ups as well as push-ups, explosive power by the vertical jump and flexibility by the modified sit-and-reach test. The females had a higher relative percent total body fat with moderate effect (p < 0.001, eta2 = 0.566) while males had more lean body mass (p = 0.005) but this difference was small (eta2 = 0.399). There was no difference between males and females in right (p = 0.355, eta2 = 0.054) and left leg flexibility (p = 0.142, eta2 = 0.130). The males jumped higher than the females (p = 0.001) but the effect was small (eta2 = 0.476). The difference became smaller when scaled for height (m2): 17.39 ± 3.07 em for males and 14.25 ± 1.66 em for females (p = 0.020, eta2 = 0.331). The theoretical exponent for height controlled for the effect of the body size variable relative to the vertical jump in the males (r =- 0.08, p = 0.864) but not in the females (r = -0.71, p = 0.033). When the vertical jump was expressed in terms of LBM (kg0 · 67 ), the difference between males and females disappeared (p = 0.179, eta2 = 0.125): 3.52 ± 0.88 em versus 3.06 ± 0.41 em. The theoretical exponent for height controlled for the effect of the body size variable relative to the vertical jump in the males (r = -0.66, p = 0.1 09) but not in the females (r = -0.83, p = 0.006). There was no difference between males and females in push-ups (p = 0.244. ES = 0.466) as well as in the sit-ups (p = 0.809, ES = 0.127). The males had a higher aerobic endurance (p = 0.003, eta2 = 0.453). Male taekwondo-in have higher aerobic endurance, higher explosive power and less percent body fat.
- PublicationPrevalence of needle stick injuries among nursing students from school of Health Sciences Universiti Sains Malaysia.(2009-04)Ibrahim, Nor Akmal MasorNursing students face the threat of needle stick injuries (NSis) with the consequent risk of acquiring blood borne infections during their clinical training. The aim of this cross sectional study was to explore the prevalence ofNSis among nursing students and to find out the level of knowledge, attitude and practice regarding NSis. About 132 of nursing students were selected randomly from four batches of nursing students at School of Health Sciences, Universiti Sains Malaysia. Data were processed using SPSS version 12.0. The data were then analyzed using descriptive statistic and Pearson Chi Square to answer the research questions. The fmdings showed that the prevalence of NSis among nursing students was high with 4 7 incidences (35.6%). Only 34% of the incidences were reported by the students. The level ofknowledge regarding NSis among nursing student was high (M = 45.23, SD = 2.73). Level of attitude regarding NSis was at the positive level of attitude (M = 3.91, SD = .49). Level of practice regarding NSis was at the moderate level (M = 2.76, SD = .31 ). There were no statistically significant associations between the level of knowledge, attitude and practice with the prevalence of NSis among the nursing students (p < .OS). Overall, this study showed that nursing students are at high risk of needle stick injmy during their clinical activities. Therefore, some preventive strategies should be taken by the university and hospitals to avoid the occurrence of these problems. Nursing students also must be aware and always perfonn the correct nursing practice by following the guidelines of universal precaution effectively.
- PublicationQualitative phytochemical analysis on psidium guajava leaves extracts and its inhibition effects on human osteosarcoma cells.(2009)Ruslan, Nor FarhanaThe world of modem medicine is now beginning to accept the use of botanicals once they are scientifically validated. One example is the Psidium guajava or commonly known as guava. The leaves of Psidium guajava (PG) is very well known for its vast traditional uses and pwposes. One of the areas in concern is the potential chemopreventive activities. The objective of this study is to perform qualitative phytochemical evaluation on the extracts of PG leaves (methanol extract, petroleum ether extract and water extract) and also to determine the inhibition concentration (IC50) of the extract on the osteosarcoma cell lines (CRL 1543). Leaves collected from one of the local household in the Kelantan state is processed, and extracted using three different types of solvents, namely petroleum ether (PE), methanol and water. The qualitative phytochemical analysis done on the extracts showed that the methanol and water extracts contain alkaloids, tannins and saponms. The petroleum ether (PE) extract on the other hand contains only alkaloids and terpenoid. The methanolic extract showed significant growth reduction activity in CRL 1543 and the IC50 concentration of the methanolic extract on CRL 1543 is approximately 50 ug/ml (p value< 0.05)
- PublicationThe study of transgene intergration hotspots in mammalia expression system(2015)Shamsuddin, ShaharumMammalian expression system is very crucial for the production of proteins in numerous scientific and commercial areas as they are able to perform proper protein folding as compared to other hosts. Matrix attachment regions (MARs) are concentrated with transcription factor binding sites and have a strong effect on the level of expression of transgenes, thus highly studied to increase protein production. In this study, an expression vector carrying green fluorescent protein was transfected into mouse myeloma NSO cell line in order to allow random integration of the vector into the host genome. Conceptually, if the vector is integrated into a transcriptionally active site within the genome, green fluorescent protein will be highly or constitutively expressed. Transfected cells were then subjected to sorting in order to select the high producing cells using ClonePix FL (CPL). CPL is an automated high-throughput clone selection instrument. DNA sequences in the genome flanking the integrated vector in the high producing cells were subsequently retrieved using ‘genome walking’ method. Following this, the flanking regions were sequenced and characterized using bioinformatics tools. Functional analysis was carried out by entering the sequences into three different programs (SMARtest, MARfmder and MARscan) to determine the presence of Matrix Attachment Regions. In addition, biochemical characterization was also done using Chromatin Immunoprecipitation (ChIP) Assay. As a result, putative MAR elements were identified in mammalian expression system with different lengths of MAR regions between the three algorithms. MARJnndiii proteins that were used in CMP assay were also shown to interact with the mouse
