Abstract
A recombinant clone designated pCYS9 was isolated from as. typhi genomic library by
screening with an :lnti-sel'ull1 ;lgainst S. tYflhi antigens.
Characterization of the pCYS9 recombinant clone showee!thal the insert DNA is 2,0 ISbp
in size. Different fragments of the insert were usee! to probe chromosomal DNA of
various SO/lilollci/Of species andorgallisills of other gellus. All C) probes delecleel S.
t)'phi aile! 5 other S(t/IJ1?lIe/loc species tested whereas none or the 9 nOIl-.,,'({11I101l(,//({C
organisms were detected by allY or the probes. These results showed th:lt the insert or
pCYS9 is specific for S(fIIlIOI/(~II(/e.
Sequencing of the pCYS9 insert DNA W;IS then c;IITiecl oul alld the complete sequellce
was acquired. Comparison of the insert DNA sequence witl} DNA sequences in the
Microgcnie (Beckmall, USA) progrnlllllle databank and other databanks such ;IS Gellhank
;lIlci [MI3L using llle BLAST programl1le through the Nation:ll Center or Biotechnology
Information (NCBI) E-Mail Server showed thal bOlh ends 0(' the inscrl DNA h:1S
significant homology wilh a few reported DNA sequences. However, tile middle region
of the insert DNA between resiclues 840br to I ,200bp has no significant hOlllology wilh
any reported sequences.
Comparison :lIttle :llllillO ;lcid level clelected ;1 LlyYtJ - 50% hOl1lology between Ihe illscrt -
ami a few cobra cyloloxin proteills between residues 850-940bp lIsing the Microgcnie·
programille (Beckman, USA).
The results obtained suggest that this 2.0kb DNA fragment has the potenLial [0 be
developed ;\s a specific probe for Salmollelloe.
