Malaria is a blood-borne infectious disease that continues to pose a significant threat to both children and adults, particularly in tropical and subtropical regions of the world. In this research, the phytochemical analysis of the ethyl acetate (etoac) bark extract of two diospyros (d.) genus plants consisting of d. Adenophora and d. Lanceifolia was carried out. A total of nine (9) compounds were successfully isolated from both plants using column chromatography (cc) and thin layer chromatography (tlc) methods. The compounds were identified as five (5) pentacyclic triterpenes, one (1) steroid, two (2) fatty acids and one (1) phenolic compound. Three compounds were isolated from the etoac bark extract of d. Adenophora, including lupenone (50), lupeol (46) and betulin (33). Besides, the chromatographic isolation of the etoac bark extract of d. Lanceifolia has led to the isolation of eight compounds such as lupeol (46), betulin (33), β-sitosterol (47), oleic acid (58), α-amyrin acetate (59) glyceryl trilinoleate (60), β-amyrin (61) and shinanolone (62). Following this, the major isolated compounds of lupeol (46) and betulin (33) from the etoac extracts of two plants were combined for hemisynthesis via acylation, hybrid reaction and oxidation reaction, which led to the synthesis of twenty-three (23) derivative compounds.
