Abstract
Artcmisinin (qi-is) and its derivatives are the most pronusing compounds against multidrug resistant falciparum malaria. Artemethcr (arm) and artecther (are) were found to be more effective than their parent compound, qhs. The objectives of the study were to improve the sensitivity of the analytical methods for the quantification of arm and are, together with their majur metabolite, dihydroartemisinin (dqhs) in plasma, and to study the pharmacokinetics of arm in healthy volunteers and in patients with uncomplicated falciparum malaria. The present study has developed a rapid, selective, reproducible and specific high performance liquid chromatography with electrochemical detector for the determination of arm and dqi-is with the sensitivity limit in plasma of 2.5 and 1.25 ng/ml respectively. Through minor modifications, the analytical procedure was extended to the analysis of are in plasma with a sensitivity limit of 5.0 ng/ml. The method was succesfully applied in clinical pharmacological studies of arm in human.
