dc.description.abstract | A preliminary study on the antimicrobial activities of six selected plants was carried out. Four of them were wild plants namely Elettariopsis triloba, T;phonium trilobatum, Homalomena rubra and Curculigo latifolia. One of them was an epiphyte, that was Drymoglossum pilose!!oides and the other was a traditional medicinal plants, Orthosiphon grandiflorus. Lipid and aqueous solvents were used for the extraction. :\ntimicrobial activity test on 21 pathogenic microorganisms showed that none of the aqueous extracts exhibit any antimicrobial activity while all the ethanol extracts showed significant activities. Most of the ethanol extraction showed broad spectrum of activities on gram positive and negative bacteria but not on yeast and fungi. The ethanol extract of Drymoglossum piloselloides and Orthosiphon grandiflorus were selected for the subsequent experiments because they produced bigger inhibition zones. Extraction utilizing hexane and methanol were conducted and the extracts were used for the subsequent experiments. All the hexane extracts except Orthosiphon grandiflorus leaves only showed effect on two certain fungi species but none on bacteria and yeast. Screening results for methanol extracts indicated that both Orthosiphon grandiflorus leaves and stem had broad anti bacterial activities but not anti yeast and anti fungal activities. XXIII The MIC and MBC determination test for selected extracts and pathogens were pt:rformed to determine the lowest concentration to inhibit and exterminate pathogens. Orthosiphon grandiflorus methanol leave extract showed the lowest MIC value of 3.12 rng/ml against B. cereus where as Drymoglossum piloselloides, the methanol extract of leave portion showed the lowest MIC value of 6.25 mg/ml against K. pneumoniae. (}rthosiphon grandiflorus methanol leave extract also showed the lowest MBC value of !2.5 mg/ml against K. pneumoniae where as for Drymoglossum piloselloides, the methanol extract of leave portion showed the lowest MBC value of 12.5 mg/ml against K pneumoniae. Profile tests showed that these extracts were succeeded to influence the pathogen growth in lower concentration from the MIC values and the effect was similar at all pathogen life phases which supported by the observation from SEM and TEM. Results from the pathogen inhibition tests against the extract fractions found that the inhibition effects showed by the ethanol extract of Drymoglossum piloselloides root was sinergistic while Orthosiphon grandiflorus stem methanol extract showed that the inhibition effect was caused by the presence of specific compound in the extract. All the selected extracts also showed toxic effect on brine shrimp. | en_US |
