Abstract
Pathogenicity tests were carried out on oil palm seedlings and germinated seeds using monokaryotic and dikaryotic mycelia in order to investigate the role of dikaryotization in conferring pathogenicity. The results showed that monokaryotic isolates were non-pathogenic while reconstituted dikaryotic mycelia can initiate basal stem rot infection. The inoculation method of placing germinated oil palm seeds in direct contact with rubberwood blocks well-colonized with dikaryotic mycelia gave faster and consistent results as the disease symptoms started to appear four months after inoculation. Mating compatibility tests were carried out using Ganoderma monokaryon tester strains to study the biological relationship of G.boninense from oil palm and Ganoderma spp. from other palm hosts. The results showed that G. boninense crossed with other Ganoderma isolates from oil palm, including G. miniatocinctum and G. zonatum, as well as isolates from sealing wax palm, MacArthur palm and coconut. However, G. tornatum did not cross, which showed that it is not conspesific with all the other Ganoderma isolates from other palm hosts. Molecular analyses using PCR-RFLP of ITS1-5.8S-ITS2 showed that different restriction patterns were obtained from isolates from different hosts. UPGMA cluster analysis of the restriction patterns showed that Ganoderma isolates from various palms and hard wood (tea and rubber) were clustered separately. Therefore, PCRXV RFLP analysis of ITS1-5.8S-ITS2 strongly supports mating data in species delimitation of Ganoderma.
