dc.description.abstractFusarium subglutinans is a fungus classified in the phylum Ascomycota and can caused several economically important plant diseases. Fusarium subglutinans is one of the species traditionally placed in section Liseola. Species within this section are highly variable and difficult to differentiate from each other only by using morphological identification technique. This study was conducted to identify and characterize suspected F. subglutinans isolates deposited at the Fusarium Culture Collection, School of Biological Sciences, USM by using several taxonomic i.e. morphological, biological (mating population), and molecular approaches. A total of 63 isolates of suspected F. subglutinans were obtained from the Fusarium Culture Collection and results of the morphological identification showed 43 isolates identified as F. subglutinans and 20 isolates as F. sacchari. In mating population studies, the isolates of F. subglutinans repeatedly failed to cross with standard tester strains of that carry MAT-B and MAT-E alleles. Therefore, molecular approach of translation elongation factor-1α gene (TEF-1α) sequence was applied to further identify and characterize the F. subglutinans isolates. From the phylogenetic analysis, three species of Fusarium i.e. F. subglutinans, F. sacchari, and F. concentricum were identified. Phylogenetic trees of neighbor-joining (NJ) and maximum likelihood (ML) clustered the isolates of the same species in the same clade. Polymerase chain reaction of restriction fragment length polymorphism (PCR-RFLP) of intergenic spacer region (IGS) using six restriction enzymes, AluI, Hin6I, TaqI, Bsu15I, XhoI and EcoRI were used to further characterize the isolates. High levels of variability were obtained among the F. subglutinans, F. sacchari, and F. concentricum isolates. The dendrogram tree of UPGMA cluster analysis showed that F. subglutinans, F. sacchari, and F. concentricum were clustered according to the species. F. concentricum is a new record in Malaysia deposited in the Fusarium Culture Collection.en_US
