Abstract
Elite cell lines of misai kucing ( Orthosiphon stamineus Benth) were selected from in vitro plantlets derived from different locations based on plantlets height that can produce the optimum rosmarinic acid. Three cell lines derived from mother plants from Air !tam and Butterworth and two cell lines derived from Relau were selected based on their height. Cell suspension cultures derived from fast growing in vitro plantlets from Air Itam produced the highest content of rosmarinic acid (5.0%) after 12 days of culture. The selected cell lines grew well in 100 ml to 1000 ml Erlenmeyer flasks contained culture medium 30% of the flasks volume. Elicitors were applied to the selected cell lines and the in vitro plantlets of misai kucing to enhance the production of rosmarinic acid. Only 45 g/1 of sucrose added into the liquid cell suspension culture medium (MS + 1.0 mg/1 2,4-D and 1.0 mg/1 NAA) significantly enhanced the highest production of the rosmarinic acid (5.5%) compared to the control (4.9%). The free radical scavenging activity and total phenolic compound in the cells increased after the cells had been treated with various type of elicitors. Treatment with 1.50 g/1 yeast extract, 150 mg/1 chitosan, 2.0 g/1 sodium chloride and 45 g/1 sucrose successfully increased the free radical scavenging activity of the misai kucing cells. Meanwhile, the addition of 1.50 g/1 yeast extract, 100 mg/1 and 150 mg/1 chitosan and 45 g/1 sucrose into the culture medium increased the total phenolic compound in the misai kucing cells. Application of 4.0 g/1 yeast extract, 60 mg/1, 120 mg/1 and 240 mg/1 chitosan and 1.0 g/1 and 4.0 g/1 casein hydrolisate successfully increased the production of rosmarinic acid in the in vitro plantlets of misai kucing.
