Abstract
A research was conducted to study the antimicrobial effects of tannin from barks of bakau minyak, Rhizophora apiculata Blume. Three types of tannin had been extracted from barks of Rhizophora Apiculata Blume, namely mixed tannin, hydrolysable tannin and condensed tannin. Screening using with disc diffusion assay showed that the hydrolysable tannin of Rhizophora· apiculata Blume was more effective in antimicrobial activity compared to mixed and condensed tannins. In this study, special attention was given to hydrolysable tannin of Rhizophora apiculata Blume in order to find out its ability as antimicrobial agent on tested microorganisms. . .. The results of me. study revealed that hydrolysable tannin of Rhizophora apiculata Blume exhibited significant antibacterial and antiyeast activities but not antifungal activity. The minimum inhibitory concentration, (MIC) and the minimum lethal concentration (MLC), for hydrolysable tannin of Rhizophora apiculata Blume had been determined for the selected test microorganisms. The MIC value for hydrolysable tannin of Rhizophora apiculata Blume was 6.25 mglml and minimum yeastocidal concentration (MYC) value was 12.5 mg/mt for Candida albicans. The hydrolysable tannin of Rhizophora apiculata Blume at halfMIC, MIC and two times MIC concentrations can inhibit the log phase of the yeast growth, Candida albicans. 11vlicroscopic studies showed that hydrolysabk tannin of Rhi::.ophoro apiculma Blume causes some morpholot.'Y changes in the treated cells, ( ·allllidaalhiccms. In ( ·cmdiclo albicans cells, it causes abnonnal budding and dcfom1ation of the cell wall. ~ - The cytotoxicity test for hydrolysable tannin of Uhi::.oplwra apiculata Blume was carried out using brine shrimp lethality assay and HepG2 cancer cells. 1t was found that hydrolysable tannin of Rhi::ophora apiculato Blume showed significant values of lethality concentration of 50% (LC;n). The hydrolysable tannin which extracted from Rhizophora apiculata Blume also shows sib'llificant cytotoxicity effects on the HepG2 cancer cells. It was found that the number of survival HepG2 cancer cells became less as the concentration of the hydrolysable tannin of Rhizophora apiculata Blume increased. The separation of the compounds of the hydrolysable tannin of Rhi::ophora apiculata .\ .. Bh.une was done by using thin layer and column chromatography techniques. The antiyeast activity test was performed afterwards. The antagonism effects were shown by the hydrolysable tannin of Rhizophora apiculata Blume. This confinned since the separating components of the hydrolysable tannin of Rhi::ophora apiculata Blume showed a significant effect compared to the hydrolysable tannin. Through the highperformance liquid chromatography (HPLC) method, the main component of the hydrolysab1e tannin of Rhizophora apiculata Blume which react as antiyeast (Candida albicans) was confirmed as gallic acid.
