Abstract
The Asian sea bass, Lates calcarifer is a popular aquaculture fish and has a
high economic value in Malaysia. The lack of effective broodstocks for sea
bass breeding program cause problems for the management of aquaculture
populations. Extensive knowledge and data of genetic variations are
important to have successful cultivation program and increase the
aquaculture productivity. This study involves the identification of the Asian
sea bass broodstocks using microsatellite DNA markers and examines the
level of genetic variation of Asian sea bass in Malaysia using mitochondrial
DNA control region (mtDNA CR) sequencing and microsatellite DNA
markers. A total of 20 microsatellite loci were used for DNA fingerprinting of
parent and progeny in the identification of the parents to their progeny in a
mass breeding program. Simulations based on allele frequency dc;Jta showed
five loci were able to assigned 97% of progeny to their respective parents.
All 20 microsatellite loci used in this study were polymorphic (PIC = 0.52). A
total of 59 haplotypes were identified using the sequences of mtDNA CR.
This value can be considered high since haplotype diversity (h) value range
from 0.9286 to 1.000). Pairwise genetic differences between populations
(Fsi = 0.0407-0.1139 microsatellite data; 0.023-0.0597, mtDNA CR data) is at
low to moderate levels. Phylogenetic analysis using the neighbour-joining
(NJ), maximum likelihood (ML), and the maximum parsimony (MP) trees
showed similar topologies. The population of Sabah was placed in different
cluster to those of Johore and Terengganu populations with 72% (NJ), 100%
(MP) and 96% (ML) bootstrap values respectively, and the placement was
also supported by the analysis "Factorial Correspondence" (FCA) plot using
microsatellite data.
