dc.description.abstract | Six Phyllanthus species in Malaysia namely P. columnaris, P. debilis, P. emblica, P. myrtifolius, P. reticulatus and P. urinaria were selected in this study. The aims were to investigate the antimicrobial activities, to conduct phytochemical screening, toxicity test and secondary metabolite analysis in P. reticulatus as anticandidal agent. Screening for the antimicrobial activity using 18 extracts from six Phyllanthus species against eight types of bacteria, yeast and three types of fungi showed that 80% methanol extract of P. reticulatus exhibited the best activity. The highest inhibition zone of 29.75 ± 0.35 mm was exhibited when the extract was applied against C. albicans. MIC50 and MI40 values of the extract against C. albicans were 0.312 mg/ml and 5 mg/ml respectively. Hence, further study was only focused on secondary metabolite analysis in 80% methanol extract of P. reticulatus. Fractination on the extract using one dimention paper chromatography produced 12 fractions. None of the 12 fractions showed anticandidal activity compared to the crude extract. This means the extract showed inhibition when all bioactive compounds generate sinergisme. Futhermore phytochemical screening on the extract showed it contained the highest amount of flavonoid but no terpenoid was detected. The SEM study done on C. albicans showed various changes on structure and morphology of the treated yeast cells leading to cell lysis and death. The toxicity test of the extract that was conducted using brine shrimp lethality assay showed no toxicity with the LCso values of 4.36 and 1.39 mg/ml. respectively for both acute and chronic toxicities. It can be concluded that, the 80% methanol extract of P. reticulatus contains bioactive compound from the flavonoid group that have anti C. albicans activity with low toxicity. | en_US |
