Abstract
Optimization of cultural conditions and medium compositions for xylanase production by free and immobilized cells of P. chrysosporium were carried out in a shake flask system. The optimized cultural conditions. with the cultivation temperature of 30°C, agitation speed at 150 rpm, inoculum size .. ::.f 6 x 106 spores/ml and initial medium pH of 5.0 yielded about 3.5 IU/ml of xylanase activity and 0.52 gil of fungal biomass after six days of cultivation. The optimization of =tedium composition was also carried out by selection of better carbon, nitrogen and inducer sources for enhancing the xylanase production .. The optimized medium composition. which consisted of 0.1 0% (w/v) cellulose, 0.1% (w/v) ammonium chloride and 0.1 ~ t (wi\") xylan as carbon, nitrogen and inducer sources, respectively, produced about 17.8 :-.J/ml of xylanase activity and about 0.76 gil of the fungal biomass. In order to enhance the production. of xylanase and to protect the fungal cells from abrasion and shear forces during egitation in the optimized cultivation medium, the immobilization technique using scouring mesh cubes and calcium alginate beads as support materials were performed. The maximum xylanase production produced by the scouring mesh 'and calcium alginate immobilized cells were about 80.2 IU/ml and 32.4 IU/mI, respectively. There were abc·::! 400% and 180% increment of xylanase production by scouring mesh and calcium alginate immobilized cells, respectively, compared to free cells. The scouring mesh irnmobilized cells of P. chrysosporium exhibited a significant of two repeated run. The immobilized cells of P. chrysosporium in scouring mesh cubes was then cultivated in a 2.0 L stirred tank fementer system for 10 days. The results exhibited the xylanase production of about 159.3 IU/ml after 8 days of cultivation in an optimized medium with 150 pieces of 0.75 cm3 scouring mesh cubes, agitation speed of 150 rpm, aeration rate of 1.0 vvrn and inoculum size of 2.5% (w/v) of 6 x 106 spores/ml.
