Abstract
A morphological and molecular genetic study was conducted on guava cultivars obtained from various parts in Peninsular Malaysia. This is the first documentation to determine the similarities, differences and relationships between each guava cultivars utilizing morphological and molecular techniques. The guava cultivars investigated were Skg (seeded), KbsD (seeded), Batu (seeded), KbG (seeded), SdB (seeded), Aiss (seedless), Epal (seedless), Sds (seedless) and Bulat (seedless). A total of 180 samples consisting of 9 guava cultivars were used for physical morphometric measurement. All of the 6 physical morphological characters that use are namely MSp (mesocarp), EDp (endocarp), Brt (weight), Lbr (leaf Lenght2), Pjg 1 (leaf Lenght1 ), and Dmt (diameter). These physical morphometric measurement are use for supported the genetic result from Random Amplified polymorphic DNA (RAPD), where all 180 of the same plant samples were used. RAPD analyses were carried out using 5 different primers, OPB-04, OPB-18, OPF-03, OPF-09 and OPG-1 0, which produce a total of 68 polymorphic bands. Neighbor-Joining (NJ) cluster analysis generated a dendrogram, which successfully divided guava cultivars into nine clusters. More detailed studies of the physical characteristics and genetic population studies are suggested to have a better understanding of guava similarities and differences between cultivars. The differences of guava cultivars shown in molecular analyses can also be seen in their physical character more clearly.
