Abstract
The yield of fermentable sugars by saccharification of palm press fibre (PPF)
was found to be markedly higher (58% conversion) within 48 hrs of soaking PPF in
alkali (!.OM, 2 hrs} than that boiled with the same amount of alkali (47"/o
r·'conversion). Soaking conditions were optimised (I.SM, 4 hrs) which resulted in 76%
increase in conversion which is a further increase of I8%. An increase in conversion
of upto 90% was only possible with the addition· of two enzymes, cellobiase and
cellulase, during the hydrolysis of treated PPF carried out under optimum conditions.
Enzymic hydrolysis, therefore produced higher yield than that of acid, the
maximum conversion of which was about 53% under reflux at I 00°C with optimised
acid concentration of I6% (w/w), PPF size of 0.32 mm, acid-substrate ratio of I: IS
and the reaction period of 4 hrs.
The cellulosic hydrolysate containing reducing sugars was then subjected to
fermentation by Brevibacterium lactofermentum for glutamic acid production. The
optimum parameters obtained from the fermentation of the pure glucose such as pH
of 7.5, temperature of 30°C, biotin concentration of lOJ,.t.g/1 and agitation at 200 rpm
were employed.
