dc.description.abstractErythricium salmonicolor is a basidiomycetes fungus associated with pink disease affecting rubber tree (Hevea brasiliensis). Erythricium salmonicolor infection may cause poor latex production. The objectives of this study are to identify proteomic profiles and biomarkers in rubber tree related to E. salmonicolor infection. Using proteomic approach, the number of proteins that were identified from fractionated proteins of latex from E. salmonicolor infected and healthy rubber tree PB 350 were 17 and seven expressed proteins, respectively. The presumed functions of the proteins identified were related to antifungal activity, energy metabolism and antioxidation. An example of these proteins included protease inhibitor HPI that involved in defense against pathogens. Qualitative proteomic analysis and quantitative gene expression using quantitative real-time PCR showed the down regulation of specific group of genes involved in energy metabolism and transporter. Moreover, the data showed simultaneous activation of several defense and stress responsive genes expressed in reactive oxygen species (ROS) scavenging mechanism. A major finding was the transcription of E3 ubiquitin protein gene that was important in facilitating protein degradation was found up-regulated on E. salmonicolor infected rubber tree. However, down regulated of major latex allergen of hev b5 on mRNA of E. salmonicolor infected rubber tree could not be identified at the proteomic level suggesting that the expression of a gene and its protein was complicated by post-transcriptional and translational regulation. Understanding the role of the genes and proteins that are involved in the molecular and biochemical processes during Hevea-Erythricium interaction process may provide insight for developing targeted control measures and novel methods of disease management.en_US
