dc.description.abstractLeptospirosis is one of the emerging diseases of both humans and animals worldwide, particularly in the tropics. It is a major public health problem due to the lack of diagnostic facilities for early detection which is key in preventing serious multi-organ complications. Although culture and microscopic agglutination test (MAT) are gold standard, they require time and expertise and are not useful for early diagnosis. Thus this study was performed to identify antigenic components of Leptospira for sensitive and specific serodiagnosis of the disease. Proteomic approach was used for extraction, separation and identification of leptospiral antigenic components that reacted specifically with serum samples of patients with confirmed acute leptospirosis. Two pathogenic leptospira serogroups prevalent in Malaysia namely Icterohaemorrhagiae (L44) and Javanica (L55) were used for antigen preparations. A total of 93 serum samples were divided into three categories namely leptospirosis, non-leptospirosis and healthy control sera. Several solubilizing agents such as acidic glycine, TritonX-114, urea, thiourea, Tris and CHAPS were used in five antigen preparations to maximize the number of antigenic epitopes; while SDS-PAGE, two-dimensional electrophoresis (2-DE), and Western blot were performed to analyse the immunoreactivity profiles.en_US
