Abstract
Centella asiatica has been identified to reduce the declining cognitive function in studies done by ethnopharmacological scientists. In this study, asiatic acid from triterpenoid groups isolated from C. asiatica was examined for in vitro thin layer chromatography (TLC) bioautographic acetylcholinesterase inhibitory enzyme activity, in vivo psycopharmacology of excitatory post synaptic potential (EPSP) on the rat hippocampus and in vivo learning and memory potential. The mechanisms of the blood pressure with locomotors performances were also performed. Asiatic acid showed an effect on TLC bioautographic acetylcholinesterase inhibitory activity with minimal inhibition concentration which was 0.125 μg. Asiatic acid had selectively reacted to gamma aminobutyric acid receptor B (GABA B) agonist for in vivo psycopharmacology of EPSP. Then the inhibitory concentration of GABA B was plotted with half maximal inhibitory concentration, of 14 μM and Hill-Coefficient 0.9828. The in vivo learning and memory potential were performed using passive and active avoidance devices. Administration of 30 mg/kg asiatic acid facilitated the passive avoidance on memory but had no effect on learning in active avoidance. Study on the effect of blood pressure with locomotors performances showed that 10 and 30 mg/kg asiatic acid reduced blood pressure and there was no sedative effect on locomotor performance.
