| dc.description.abstract | This research was aimed at the development of novel microsatellite markers for C. striata, characterization of genetic diversity, population structure and phylogenetic relationships among wild populations inferred from mitochondrial DNA and newly derived microsatellite DNA markers. It also involved the investigation on the phylogeography aspects that had influenced current genetic distribution at macro as well as micro scale levels.. A recommendation on the management strategies (stock enhancement and conservation) and the potential candidates for broodstock selection was put forth. Mitochondrial and genotypic data consistently revealed moderate genetic diversity among all the wild C. striata populations though a wide range of variation was observed. In general, this suggested independent evolutionary history for each population studied. High genetic structuring among populations was typical of freshwater species with limited dispersal ability. However, mild gene flow could occurred between adjacent populations. Three distinct genetic lineages were apparent which discriminated the northwest Peninsular Malaysia, southeast Peninsular Malaysia and Malaysian Borneo from each other. However, an ancient anthropogenic stock translocation from Peninsular Malaysia to Malaysian Borneo was detected as inferred from both mitochondrial and microsatellite DNA data. Furthermore, haplotype sharing between the east Peninsular Malaysia and Sumatra populations revealed extraordinary migration ability of C. striata (>500km). Fine scale study in Perak revealed upstream refugia found at three different river systems but they were not differentiated from each other. | en_US |
