Abstract
Eurycoma longifolia or Tongkat Ali is used traditionally in the trea!~ent of malaria,
ulcer, fatigue and fever. This plant has also been claimed to have aphrodisiac property
in men. Concurrent use of Eurycoma longifolia with modem drugs may have the
potential to cause herb-drug interactions. The objective of this study is to investigate
the in vitro and in vivo effect of 22 Eurycoma longifolia-containing products and 2 pure
compounds on aminopyrine metabolism under the influence of disease (diabetes),
gender and age and to elucidate the molecular mechanism of the action of these
products and pure compounds on aminopyrine N-demethylase (AP N-Dase) activity.
From the study, we found that all products have the potential to influence aminopyrine
metabolism by either increasing or decreasing AP N-Dase activity. These products also
have the ability to alter the effects of diabetes, gender and age on aminopyrine
metabolism. Euryco, both in vitro and in vivo, caused significant increment in AP NDase
activity. This suggests that Euryco has direct effects on liver AP N-Dase activity.
Under the influence of disease (diabetes), gender and age, we found that the
aminopyrine N-demethylase activity in vitro was different from in vivo. This suggests
that, interactions occur between Euryco and the physiological system of the body and
these interactions are dependent on disease (diabetes), gender and age. In the molecular
mechanism study, we found that the action of Eurycoma longifolia-containing products
on AP N-Dase activity involved different second messenger pathways, including the
tyrosine kinase pathway, the calmodulin-dependent protein kinase pathway and the
pathways mediated by protein kinase A and protein kinase G.
