Abstract
Pseudomonas aeruginosa (USM-AR2) culture was proven may facilitate distillation process without reducing the quality of petroleum distillate obtained. This strain yields 10 giL of biomass in shake flask batch culture experiments, an increase of 20-fold, after 120 hours cultivation, using 2% (v/v) inoculum, cultured in 100 mL medium at an initial pH 5 at room temperature (27 °C) in the orbital shaker, agitated at 200 rpm. A suitable medium has been formulated, containing 5% (v/v) diesel, 0.6% (w/v) yeast extract, 0.05% (w/v) MgS04 and 0.05% (v/v) Tween 80. Biosurfactant activity (rhamnolipid) was detected by the orcinol assay, drop collapse test, surface tension, and viscosity measurement of oil. Treatment of crude oil TK-57S and Angsi with the improved culture of P. aeruginosa (USM-AR2) resulted in 2 to 3- fold decreases of distillation times. No obvious difference was observed among treatment with cell pellet, supernatant or the culture, suggesting that the biosurfactant is extracellular and cell bound. Batch culture experiments in 1 L working volume bioreactor gave a biomass of 10 giL achieved after 72 hours incubation. Then, fixed volume pulse feeding strategy with diesel oil as substrates resulted in 12 giL of cell biomass and 3.2 giL ofrhamnolipid achieved in shorter time, after 42 hours cultivation.
