dc.description.abstract | The production of aseptic tissue from Zingiberaceae plant could be increased by sterilising the explant tissue with I 00 mg/1 mercury cloride for five minutes, followed by Clorox® I5% for I5 minutes in the first stage and Clorox® I 00/o for I 0 minutes in the second stage. Zingiber officinale explants could produce the most number of multiple shoots, 9.8 shoots per explant, when cultured in Murashige and · Skoog (MS) medium (I962) supplemented with 2.0 mgll Benzyladenine (BA) and 2.0 mgll Indole Butyric Acid (IDA). This optimum medium was also found to be suitable for in vitro . propagation of other Zingiberaceae species namely Curcuma domestica, Curcuma zedoaria, Kaempferia galanga, Alpinia galanga and A/pinia conchigera. All these species could produce multiple shoots when cultured in this medium. For Zingiber officinale, Kaempferia galanga, Alpinia galanga and Alpinia conchigera. number of multiple shoots . formed in solid proliferative medium and liquid proliferative medium was not significantly different while Curcuma domestica and Curcuma .zedOaria produced more multiple shoots in liquid medium than solid medium. The suitable period of subculturing for Zingiber officinale, Curcuma domestica, Curcuma zedoaria and A/pinia conchigera was four weeks while the most suitable subculturing period for Kaempferia ga/anga and Alpinia galanga was eight weeks. Continuously subculturing of these species caused a decrease in the number of multiple shoots formed. After acclimatization in the growth chamber for two weeks, 77-90% of the in vitro Zingiberaceae plantlets survived and all these plantlets grew well in the field. | en_US |
