dc.description.abstract | Detection and identification of phytopathogenic bacteria infecting Artocarpus heterophyllus in this study consist of phenotype, biochemical and molecular characteristics. Through observations, the diseased plants showed symptoms of yellowing and wilting on leaves. In more severe cases, canker developed on larger branch, which finally lead to the death of the whole tree. Two types of bacteria were successfully isolated from diseased tissues taken from leaves, twigs and barks. Based on the pathogenicity tests done on tobacco leaves and jack:fruit shoots, only one group of bacteria were able to caused chlorosis to tobacco leaves and caused a fireburning character to jack:fruit shoots. The other group of bacteria was later classified as saprophyte as no positive reactions were seen on both test plants. Through morphology test done, the first group of bacteria were gram-negative bacteria with rod-coccus shaped cells, fimbriated and occurred in short chains where these isolates were able to ferment a wide range of substrates, proves by the ability to grow on numerous semi selective agar. The strains were also capable of fermenting glucose anaerobicaly. Through APIĀ® 20E test and identification via 168 rDNA region, the strain were identified as Acinetobacter sp. Multiple alignment of 168 rDNA region of 4 other closely related species revealed that the strain were closely related to Acinetobacter calcoaceticus. | en_US |
