dc.description.abstractThe abundant of agricultural wastes can cause serious environmental problems such as air-pollution, which is resulted from the wastes burning activity that usually done by farmers after the harvesting season. In order to overcome the environmental problems, the present study was undertaken by using the agricultural wastes as the co-substrates in the SSF system for mannanase production by Aspergillus niger USM F4. The study was focused on the physical and chemical improvements of mannanase in a flask and tray systems, the mannanase purification and the characterization of the purified mannanase. Compared to the other cosubstrates (oil palm frond, rice straw and sugar cane baggase), rice husk was found as the preferable co-substrate for mannanase production. The improvement of mannanase in a flask system was carried out and the improved conditions were 5 g of solid substrate using 0.75 mm particle size of co-substrate, with ratio moisture content of 1:1 (w:v), cultivation temperature of 30°C, inoculum size of 6x106 spores/ml, static condition, with the additional of 1.6% (w/w) LBG (which act as additional carbon source and main substrate for A. niger USM F4 to produce mannanase) and 1.2% (w/w) ammonium nitrate. Under improved physical and chemical conditions in a flask system, the yield of mannanase achieved up to 68.53 U/g substrate. There was an increment of 4078.66% compared to the activity before improvement.en_US
